•Penicillin is the 1st antibiotic to be discovered•Bacetricidal due to inhibition of peptigoglycan layer breakdown  in bacterial cell wall•Resistance due to beta-lactamases, altered PBP & porin channels (Gr –ve)•Currently these are indicated in some specific indications only because of emergence of resistance•Safety profile well established except chance of hypersensitivity reactions
